爬山虎体细胞胚的发生及组织学研究 被引量:10

摘要 该文以爬山虎子叶的叶片和叶柄为试验材料 ,对爬山虎体细胞胚发生进行了研究 .结果表明 :爬山虎子叶在MS附加 6 BA 0 3mg L和NAA 0 1mg L培养基上或 1/ 2MS附加 6 BA 0 5mg L和NAA 0 1mg L培养基上都能诱导产生体细胞胚 ,子叶叶柄在 1/ 2MS附加 6 BA 0 3mg L的培养基上也产生了体细胞胚 .胚性愈伤组织转移到无植物生长调节剂的MS或B5培养基上均能发育成正常植株 .组织学观察表明 ,爬山虎体细胞胚的形成经历了球形胚、心形胚、鱼雷胚、子叶胚等几个阶段 ,逐渐发育成正常植株 ,这与合子胚的发育途径相似 .体细胞胚在发育过程中出现了连体胚、芽端畸形胚。 Somatic embryogenesis of Parthenocissus tricuspidata is studied in this paper.The results show that somatic embryos are obtained from cotyledons of P.tricuspidata induced on MS medium containing 0.3 mg/L 6--BA and 0.1 mg/L NAA or 1/2MS medium containing 0.5 mg/L 6--BA and 0.1 mg/L NAA,and from cotyledon petioles induced on 1/2MS medium containing 0.3 mg/L 6--BA.Embryonic callus cells are developed to normal seedlings when subcultured on MS or B\-5 medium without plant growth regulator.Histological observations reveal that the somatic embryos successively differentiate through globular-shaped,heart-shaped,torpedo stages and cotyledon stages,similar to the development of zygotic embryo.There are several abnormal embryos including joined embryo,abnormal-bud embryo and hyperhydric embryo,etc.in the induction stage of somatic embryos.
