白介素1β诱导骨关节病半月板生成炎性因子

摘要 目的 阐明白介素 1β(IL 1β)诱导骨关节病 (OA)的膝关节半月板一氧化氮 (NO)和前列腺素E2(PGE2 )的合成及释放。方法 取 2 2例OA患者的膝关节半月板组织 ,体外组织块培养过程中加入 0 1ng/mlIL 1β进行诱导。对半月板组织中的诱导型一氧化氮合成酶 (iNOS)的表达进行检测。定量分析诱导后的组织培养上清液中NO和PGE2 的含量。结果 所有OA患者的半月板组织中都有NO稳定生成〔(113 4 2± 9 4 8)μmol·g-1·2 4h-1〕。IL 1β诱导后 ,iNOS的表达和NO、PGE2 的生成均明显上升 (均为P <0 0 1)。结论 OA患者炎症反应中产生的IL 1β可以刺激半月板组织 ,使NO和PGE2 的生成增多 。 Objective To determine the induction of NO and PGE 2 production by IL-1β in OA knee menisci. Methods Menisci were obtained from 22 patients with OA. Tissue explants were cultured alone or with 0.1 ng/ml IL-1β in vitro. The release of NO and PGE 2 from the tissue as well as the presence of inducible nitric oxide synthase were measured. Results All menisci steadly produced NO 〔(113.42±9.48) μmol·g -1·24 h -1〕. In the presence of IL-1β, the expression of iNOS and the production of NO and PGE 2 increased significantly(P<0.01). Conclusion Menisci from patients with OA produce NO and PGE 2 in a manner which is stimulated by IL-1β. The overproduction of NO and PGE 2 may further contribute to the procedure of chondrocytes apoptosis.
