血管性痴呆大鼠海马IL-1β、TNF-α含量变化及大蒜素干预的研究 被引量:11

摘要 目的 观察血管性痴呆 (vasculardementia ,VD)大鼠海马区炎性细胞因子IL 1β、TNF α含量变化与认知功能变化的相关性 ;观察大蒜素对炎性细胞因子IL 1β、TNF α含量的影响。 方法 采用Pulsinelli 4血管阻断 ( 4 vascularocclusion ,4VO)改良法建立大鼠动物模型 ;Wistar大鼠随机分为对照组 (n =10 )、4VO组、大蒜素组 ;后两组又分为 3d、1周、4周、2个月 (n =10 ) 4个时相点 ;大鼠学习记忆能力测试采用微机控制的穿梭箱主动回避反应系统 ;IL 1β采用双抗体夹心ELISA法 ;TNF α测定采用放射免疫分析法。结果 ① 4VO组大鼠主动回避反应 (activeavoidanceresponse ,AAR)习得率显著降低 ;大蒜素治疗后AAR习得率较 4VO组显著增高 ;② 4VO组大鼠海马区IL 1β、TNF α含量于造模后 3d、1周非常显著增高 (P<0 0 1) ,大蒜素组显著低于 4VO组 (P <0 0 5 )。结论 血管性痴呆大鼠认知功能改变与大鼠海马区炎性机制改变有关 ;大蒜素可抑制炎性细胞因子IL 1β、TNF α的产生 。 Objective To observe the relationship between content changes of interleukin 1β (IL 1β), tumor necrosis factor α (TNF α) in hippocampus and cognitive lesion in vascular dementia (VD) rats and the interventional effect of garlicin on the contents of IL 1β and TNF α Methods Rat model was established by Pulsinelli's 4 vessel occlusion (4VO) technique Wistar rats were randomly divided into control group ( n =10), 4VO group, and garlicin treatment group Four time points were set in 4VO group and garlicin treatment group (3 d, 1 week, 4 weeks, and 2 months) ( n =10) Learning memory ability of rats was tested by the shuttle box system controlled by computer IL 1β was detected by ELISA, and TNF α by radioimmunoassay (RIA) technique Results Rate of active avoidance reactivity (AAR) in 4VO group was significantly decreased, but markedly improved in garlicin treatment group Contents of IL 1β and TNF α in the hippocampus of rats in 4VO group were increased markedly at 3 d and 1 week after modeling ( P <0 01), but were lower in garlicin treatment group ( P <0 05) Conclusion Changes of cognitive functions of VD rats are associated with the changes of IL 1β and TNF α in the rat hippocampus Treatment with garlicin may inhibit the excretion of IL 1β and TNF α, resulting in amelioration of cognitive functions of rats
