摘要 目的 研究脑出血后血肿周围水肿带内细胞凋亡与bcl 2表达的关系。方法 使用胶原酶法建立大鼠脑出血模型。于脑出血后 6、12、2 4h、3、7d选取血肿灶周围脑组织制成冰冻切片 ,对切片进行相应染色 :用TUNEL法观察血肿灶周围细胞凋亡 ,通过免疫组化和原位杂交方法分别从mRNA和蛋白的水平观察bcl 2的表达。对染色结果进行图像分析及统计学处理 ,分析细胞凋亡与bcl 2表达的关系。结果 脑出血 6h后血肿周围出现TUNEL阳性细胞 ,此时bcl 2的表达降低 ;之后凋亡细胞数量逐渐增加 ,至 3d达到高峰 ,此时bcl 2的表达仍处于低水平 ;3d至 7d凋亡细胞数量减少 ,而bcl 2的表达增加。结论 脑出血后血肿周围存在细胞凋亡 ,而bcl Objective To explore the relationship between the apoptosis and bcl 2 expression around the hematoma in intracerebral hemorrhage (ICH). Methods The ICH rat model was established by intracranial injection of Ⅳ collagenase. Brain tissues around the hematoma were collected at 6, 12, and 24 h, and 3 and 7 d after hemorrhage for the preparation of frozen sections for staining. Apoptosis was observed by TUNEL, and bcl 2 expression was observed by immunohistochemistry and in situ hybridization. The relationship between apoptosis and bcl 2 expression was analyzed by image analyzer and statistical methods. Results TUNEL positive cells were found around the hematoma at 6 h after ICH. At the same time, the expression of bcl 2 was low. And then, the TUNEL positive cells increased and reached the peak at 3 d after ICH. Meanwhile, the expression of bcl 2 was still very low. Then the TUNEL positive cells decreased until 7 d after ICH, but the expression of bcl 2 increased. Conclusion Apoptosis can be found around the hematoma after ICH. The decreased expression of bcl 2 is one of the most important step in ICH induced apoptosis.
