摘要
应用玻璃珠层析和密度梯度离心法从二花脸和长白各5头母猪大脑皮质中制备血脑屏障微囊,从微囊浓度、最适反应时间、离子通道、渗透压等角度分析影响血脑屏障转运卵泡抑制素的因素,获得最适工作条件,建立体外转运方法。应用该方法分析猪血脑屏障转运卵泡抑制素的动力学参数,发现二花脸猪和长白猪的Km值分别为0.17和0.07 pmol·L-1,Vmax值分别为1.15和2.27 pmol·min-1·mg-1(P>0.05)。
Vesicles of blood-brain barrier (BBB) from 5 sows in each of Erhualian and Landrace breeds were purified by beading chromatograph and density gradient centrifugation, and were applied to the transport of follicular inhibin by method in conditions of suitable vesicle concentration, reaction time, ion chunnel, osmosis, etc. The results showed that follicular inhibin could be transported by the blood-brain barrier of sows. The Km values of transport in Erhualian and Landrace breeds were 0.17 pmol·L-1 and 0.07 pmol·L-1, respectively. The Vmax values of transport were 1.15 and 2.27 pmol·min-1·mg-1 (P >0.05), respectively.
