五海丸对体外培养肺癌细胞生长及诱导细胞凋亡的影响

Experimental Study on Apoptosis and Growth on Lung Cancer Culture Cells in Vitro by Wuhai Pill(五海丸)

目的观察中药五海丸对体外培养肺癌细胞生长的影响及其诱导肺癌细胞凋亡作用。方法应用血清药理学方法,对小鼠肺腺癌LA795细胞株进行体外培养,予五海丸、环磷酰胺(CTX)、顺铂(DDP)进行干预,通过MTTI法、流式细胞仪法、DNA提取凝胶电泳法等对肺癌细胞生长及其凋亡指标进行测定。结果五海丸能够抑制体外培养肺癌细胞的生长,高剂量组杀伤率可达(59.48±9.87)%,并能够促进肺癌细胞凋亡;高剂量组凋亡指数为(27.75±4.15),与CTX合用既可以降低其增殖指数,又可增加其凋亡指数。结论五海丸能够抑制肺癌细胞生长,促进肺癌细胞凋亡,与化疗药物具有协同抑瘤作用。

Objective:To observe the effects of apoptosis and growth of lung cancer culture cells in virto by the chinese herbal medicine Wuhai Pill(五海丸). Methods:Using the method of serum pharmacology.The apoptosis and growth of lung cancer culture cells in vitro were detected with Wuhai Pill,CTX and DDP by MTT method,flow cytometry(FCM),DNA gel electrophoresis.Results:The results showed Wuhai Pill had a remarkable growth-inhibition on lung cancer cells in vitro.In high dose group the inhibitory rate was(59.48±9.87)%,and apoptosis could be promoted which apoptosis rate of cells was(27.75±4.15)%.Combining high dose group with CTX both decreased the lung cancer cells growth and increased the apoptosis rate.Conclusion:Wuhai Pill can inhibit growth and promote the apoptosis of lung cancer cell,and it can echance the effects of chemical treatment on inhibiting lung cancer.
