摘要
本文以硝酸纤维素膜(Nitrocellulose paper,NCP)作载体首次建立了检测AFP的免疫酶斑点试验(Immnue Enzyme Dot Assay,IEDA),其原理与ELISA夹心法基本相同。我们首先以AFP标准品作试验,进行方法学探索,选出最佳包被抗体浓度为100μg/ml,酶抗体稀释度为1:2000,同时发现包被过程中的封闭处理能减少非特异性反应,叠氮钠终止反应能确保显色结果稳定。试验证明,此方法特异性强,重复性好,灵敏度达1.25μg/ml。本法检测17例患者血清,其AFP含量与RIA结果相吻合。
This paper described an assay for detection of AFP with Immune Enzyme Dot Assay (IEDA) on nitrocellulose paper (NCP) . The principle and the process of this method were stmilar to those of ELISA-sandwich method. In this experiment the optimization of coating antibody concentration was 100g/ml and the optimal anti-AFP-HRP conjugate dilution was 1:2000. The non-specific stain might be avoided with 50%FCS block during coating,NaN3 was used to interrupt the further reaction. The value of AFP of 17 patients assayed on this method was approximate agreement with that assayed by RIA.
