摘要
本文比较分析了ELISA和ABC两种方法在肿瘤McAbS筛选应用中的一些利弊关系。结果表明,ELISA测出为阳性并与结肠癌细胞系反应的99.5%(198/199)孔杂交瘤细胞所分泌的抗体,经ABC法染色后是不与其癌组织反应的,其中只有0.5%(1/199)孔细胞的抗体才是这两种方法同时均能测得为阳性和是有意义的。而ABC法不仅可检出ELISA阳性的抗体,而且亦能检出ELISA漏筛为性阴、最终是有意义的理想抗体。
Some advantages and disadvantages of the two methods, an enzyme-linked immunoassay (ELISA) and an avidin-biotiny-horse-radish peroxidase complex (ABC) , in sreening of tumor monoclonal antibodies (McAbs)were compared and analysed this paper. The results indicated that 99.5% (198/199 wells) McAbs which were positive and reacted with colon carcinoma cell lines determined by the ELISA did not recognize the antigens on colon cancer tissues by the ABC. Of 0.5% (1/199 wells) McAbs to be both positive and ideal could be detected by the two assaies. It was suggested that the positive or negative McAbs which did not react with the cell lines butt could really react with the tissues assayed by the ELISA could be determined by the ABC.
