摘要
目的 :探讨电子直线加速器照射对SHG4 4细胞MMP2、ppGalNAcT2表达的影响 ,并探讨牛磺酸对此影响的防护作用。方法 :电子直线加速器 15Gy照射细胞SHG4 4作为阳性对照组 ,不照射的作为阴性对照组 ,另设三组加药组 15Gy照射 ,加药量分别为 5 0mg/L、10 0mg/L、2 0 0mg/L ,照后 12h收集。采用RT PCR法检测MMP2、ppGalNAcT2mRNA的表达差异变化。结果 :电子直线加速器照射SHG4 4细胞后MMP2上升 ,ppGalNAcT2下降 ,两者呈负相关性。而牛磺酸可使细胞在照射后与阳性对照组比MMP2下降 ,ppGalNAcT2上升 ,且与剂量相关。结论 :15Gy电子直线加速器照射促进SHG4 4细胞MMP2的表达 ,而对ppGalNAcT2有抑制作用 ;不同剂量的牛磺酸通过下调MMP2 ,上调ppGal NAclT2 ,从而抑制电离辐射引起的肿瘤细胞潜在的浸润和转移能力 ,达到生物防御作用。
Objective: To study the effect of radiation with electron linear accelerator on expressions of MMP2 and ppGalNAcT2 to SHG44 leukemia cells and Tau protection to the irradiation effect. Methods: The cells in the control group and Tau (50 mg/L、100 mg/L、200 mg/L) groups were irradiation 15Gy with electron linear accelerator.The expressions of MMP2 and ppGalNAcT2 were examined through RT-PCR within 12h. Results: The expressions of MMP2 were enhanced after irradeatiom with linear accelerator,while the ones of ppGalNAcT2 were diminished.In the Tau groups the expressions of MMP2 were diminished and the ones of ppGalNAcT2 were enhanced in turns by contrast with the control group,which correlated with dosage. Conclusion: Radiation with electron linear accelerator at a dose of 15Gy can stimulate the expressions of MMP-2,but inhibited the ones of ppGalNAcT2 in SHG44 cells.Tau inhibits more invasiveness and migration of tumor cells by the down-regulation of MMP2 expressions and the up-regulation of ppGalNAcT2 ones,so it had the biologic protection on cell levels.
