ABC法在单纯疱疹病毒感染快速检测中的初步应用

Use of ABC Method for Rapid Detection of HerPes Simpiex Virus Infection

本文在兔单纯疱疹病毒性角膜炎(HSK)模型眼研究用培养24h联合ABC法染色(C-ABC)快速检测HSV感染,并在感染HSV-1的Vero细胞制成的抗原片上比较了ABC法、PAP法、IEA和IFA在检测HSV抗原的敏感性。结果C-ABC方法检测HSV感染的相对敏感性为95.8％,特异性为100％,其阳性率与单纯培养7天结果接近,而显著大于单纯培养24h的阳性率(P<0.01)。抗原片敏感性比较表明ABC法较PAP法、IEA及IFA敏感8～64倍。提示用细胞培养联合ABC染色是一种较常规抗原检测方法更敏感、特异的HSV感染快速检测方法。

In this paper, ABC method combined with 24h culture (c-ABC) after incubated specimen into vero monolayer cell for rapid detection of herpes simplex virus (HSV) infection was reported in a experimental rabbit herpes simplex keratitis. The sensitivity of the ABC method was compared with PAP method, IEA and IFA for detection of HSV Ag. The results showed that the relative sensitivity by using C-ABC method was 95.8% (compared with 7 days vero cell culture for detection of HSVinfection), it was significantly higner than that with 24 h vero cell culture alone(P<0.01). This study also showed that ABC mehond was more sensitive and rapid than PAP, IEA and IFA methods for detection of HSV Ag.
