甲基莲心碱、红霉素对K562/A02细胞内谷胱甘肽含量的影响 被引量:9

摘要 目的 :探讨甲基莲心碱 (Nef)、红霉素 (EM)对K5 6 2 /A0 2细胞内谷胱甘肽 (GSH)含量的影响 ,及其对白血病细胞代谢解毒系统介导的多药耐药 (MDR)的逆转作用。方法 :采用生物化学DTNB法测定细胞内GSH含量。结果 :K5 6 2 /A0 2细胞内GSH含量为 (137.34± 33.10 )mg/mgprot,高于K5 6 2细胞内GSH含量 (73.38± 10 .0 2 )mg/mgprot(P<0 .0 5 ) ,且为K5 6 2细胞的 1.87倍 ;Nef10mg/L ,EM 10 0mg/L ,Nef10mg/L +EM 10 0mg/L ,维拉帕米 (VRP) 5mg/L处理 2d后K5 6 2 /A0 2细胞内GSH含量分别为 (6 9.0 1± 15 .99)mg/mgprot,(70 .5 7± 11.93)mg/mgprot,(6 5 .74± 13.37)mg/mgprot,(70 .86± 16 .16 )mg/mgprot,均较无药组含量 (137.34± 33.10 )减低 (P <0 .0 5 )。结论 :细胞内GSH含量增高是K5 6 2 /A0 2细胞产生MDR的机制之一 ;Nef,EM能使K5 6 2 /A0 2细胞内GSH含量下降可能是其逆转耐药白血病细胞MDR的机制之一。 Objective To investigate the reversal effect of multidrug resistance (MDR) mediated by glutathione (GSH) detoxifcation system in K562/A02 cell line by neferine and erythromycin. Methods Cellular GSH concentration was examined by biochemical analyses with DTNB. Results Cellular GSH concentration in K562/A02 cell line [(137.34±33.10)mg/mgprot ] was higher than that in K562 cell line [(73.38±10.02 )mg/mgprot](P<0.05),and 1.87-fold greater in K562/A02 cell line compared with that in K562 cell line. Cellular GSH concentration in K562/A02 cell line treated with Nef10 mg/L,EM100 mg/L,Nef10 mg/L+EM100 mg/L,and VRP5mg/L for 2 days [(69.01±15.99) mg/mgprot, (70.57±11.93)mg/mgprot, (65.74±13.37)mg/mgprot, and (70.86±16.16 )mg/mgprot,respectively] decreased than that in K562/A02 cell line without drugs (P<0.05). Conclusion Increase of cellular GSH concentration was one of the MDR mechanisms in K562/A02 cell line. Decrease of cellular GSH concentration may be one of the MDR reversal mechanisms in K562/A02 cell line by neferine and erythromycin.
