麝黄消瘤汤对人肝癌细胞系的抑制增殖作用研究

Anti-proliferation effect study of Shehuangxiaoliu decoction on human hepatocarinoma clone

目的 研究麝黄消瘤汤对人肝癌细胞系 (Bel- 74 0 2 )的抑制增殖作用。方法 制备含药血清 ,孵化人肝癌细胞系 (Bel- 74 0 2 ) ,用MTT比色法、倒置相差显微镜观察法及PCNA(增殖细胞核抗原 )免疫组化染色法等方法研究不同浓度的麝黄消瘤汤含药血清 ,经过不同作用时间对人肝癌细胞系的抑制增殖作用。结果 麝黄消瘤汤含药血清对人肝癌细胞系有明显杀伤作用 ,最大杀伤率达 98.4 5 % (含药血清浓度为 30 % ,孵化 72h) ;可明显降低人肝癌细胞系PCNA阳性细胞率 ,含药血清浓度 2 0 %、孵化 72h后PCNA阳性细胞率为 97.32 %。

Objective It is to study the anti-proliferation function of Shehuangxiaoliu decoction on human hepatocarinoma clone (Bel-7402).Methods Serum included with Shehuangxiaoliu decoction was produced and the Bel-7402 was incubated. Then the anti-proliferation function of the drug serum on Bel-7402 at different content and different time was studied with MTT method, assay observing method with phase-contrast microscope and PCAN (proliferating cell nuclear antigen) immuno-chemical staining method.Results Shehuangxiaoliu decoction had obvious antipersonnel function on Bel-7402. The maximal kill rate was 98.45% (the content of the drug serum was 30%, incubating for 72?h). It could obviously lower down the positive rate of PCNA Bel-7402 cell. When the content of the drug serum was 20% and incubating for 72?h, the rate was 97.32%.Conclusion Shehuangxiaoliu decoction has obvious anti-proliferation function on Bel-7402.
