摘要 目的 观察刺五加皂苷 (ASS)对缺血性脑损伤的作用。方法 采用神经元缺血性损伤模型。用流式细胞仪检测细胞凋亡率 ,用硝酸还原酶法测定细胞培养上清液中NO的含量 ,用MTT、LDH测定神经细胞活性 ,并在电镜下观察细胞形态学变化。结果 ①缺血性神经元发生凋亡 ,细胞超微结构呈现凋亡样改变 ,其凋亡率与正常对照组比较有显著性差异(P <0 0 1)。ASS能减少缺血性神经元凋亡。②缺血性神经元存活率下降、LDH释放量和NO含量升高 ,与正常对照组比较有显著性差异 (P <0 0 1)。ASS能提高神经元存活率、降低LDH释放量及NO含量。结论 一定浓度的ASS对缺血性神经元凋亡有保护作用 。 Objective To explore the effect of acanthopanax senticousus saponins (ASS) on ischemic brain damage. Methods The model of ischemic brain damage was established on cultured cortical neurons. The rate of neuronal apoptosis was measured by flow cytometer system, NO contents were determined by nitrate reductase assay and neuron viability was measured by MTT assay and release of LDH. The changes of morphology were observed by electron microscopy. Results ①Compared with the control group, ischemia could induce cultured cortical neuron apoptosis and had the rate of neuron apoptosis increased. The most striking ultrastructural changes of apoptosis were observed by electron microscopy in ischemic neurons. ASS could decrease cultured cortical neuron apoptosis induced by ischemia. ②Compared with the control group, the neuron survival decreased and the release of LDH and the NO contents increased in ischemic group (P<0 01). ASS could decrease cultured cortical neuron LDH quantity, inhibit the NO contents and increase neuron survival. Conclusions ASS plays a protective role in the cortical neuron apoptosis induced by ischemic via inhibiting the release of NO and stabilizing cell membrane.
