内皮素在蛛网膜下腔出血诱发脑血管痉挛中的作用 被引量:8

摘要 目的:探讨实验动物蛛网膜下腔出血诱发脑血管痉挛时内皮素-1(Endothelin-1,ET-1)基因在大鼠脑底动脉的表达变化及其诱发脑血管痉挛中的作用。方法:56只Wistar大鼠随机分为正常组、蛛网膜下腔出血组(2、3、7、14d)、生理盐水组和假手术组,应用逆转录-聚合酶链反应(RT-PCR),以β-actin为内参照基因,分别检测各组实验动物脑基底动脉ET-1 mRNA的表达变化。结果:蛛网膜下腔出血组,在出血2d、3d组实验动物脑基底动脉ET-1 mRNA表达较正常组明显增高,出血后7d组仍增高,14d组趋于正常。结论:ET-1参与了蛛网膜下腔出血诱发脑血管痉挛的病理生理过程,提示ET-1在蛛网膜下腔出血诱发脑血管痉挛中起着重要的作用。 To detect the expression of Endothelin-1 (ET-1) mRNA in rat cerebral basilar artery during subarachnoid hemorrhage-induced cerebral vessel spasm as well as its effect. Methods: 56 Wistar rats were randomly divided into normal、experimental、sham-operated and saline-control groups, and then the experimental group was divided into subarachnoid hemorrhage (SAH)2、3、7 and 14d groups. Reverse transcription polymerse chain reaction was used to study the alteration of Endothelin-1 mRNA level of rats cerebral basilar artery with β-actin as an inner consult during subarachnoid hemorrhage-induced cerebral vessel spasm. Results: ET-1 mRNA level of basilar artery in SAH2、3 and 7d group was higher obviously than that of normal group while that of 14d group was approximate to normal group. Conclusions: ET-1 participate in pathophysiological process of SAH-induced cerebral vessel spasm, and it suggests that ET-1 play an important role in SAH-induced cerebral vessel spasm.
