摘要
目的 :比较辛夷超临界萃取物 (XY SFE)和水蒸气蒸馏提取物 (XY WVD)对豚鼠变应性鼻炎 (AllergicRhinitis ,AR )模型的治疗作用和P物质 (SubstanceP ,SP)及其受体 (SubstancePReceptor ,SP R)mRNA表达的影响。 方法 :以卵白蛋白 (Ovalbu min ,OV)为致敏原 ,用豚鼠复制人类AR模型 ,观察症状及鼻粘膜病理状况以研究XY SFE对豚鼠AR的治疗作用 ;放免法测定鼻腔分泌物中SP浓度 ,逆转录 多聚酶链反应 (reversetranscriptivepolymerasechainreaction ,RT PCR)测定鼻粘膜中SP RmRNA含量以研究XY SFE对SP/SP R分泌及表达的影响。结果 :两种提取物均能有效改善豚鼠AR症状 ,减轻鼻粘膜病理性改变 ,调低SP/SP RmRNA分泌及表达。结论 :辛夷超临界萃取物和水蒸气蒸馏提取物对人类AR可能有类似的治疗作用 ,且其作用机制与调低SP/SP R分泌及表达有关。
Objective: Compare the effect between Xinyi(magnolia flower) XY-SFE and XY-WVD on the allergic Rhinitis(AR) model of guinea pig and the mRNA expression of Substance P(SP) and its receptor(SP-R),thus to discuss the therapy mechanism of Xinyi and the meaning of SP/SP-R expression in AR pathogeny. Method: Use ovalbumin (OV) as the sensitinogen ,use guinea pig to copy the human model,observe the symptom and pathologic condition of nasal mucosa to research on the therapeutic effect of XY-SFE on AR;use the method of radio immunity to determine the SP concentration of nasal secretion and use the reverse transcriptive polymerase chain reaction (RT-PCR) to assay the content of SP-R mRNA in mucosa to determine the effect of XY-SFE on SP/SP-R secretion and expression. Results: The two extract can both improve the guinea pig AR condition and alleviate the pathologic change of nasal mucosa,decrease the secretion and expression of SP/SP-R mRNA. Conclusion: The XY-SFE and XY-WVD extract of Xinyi may have the similar therapeutic effect on human being,its mechanism may relate to the depression of SP/SP-R secretion and expression.
