摘要
目的 研究枯否氏细胞抑制剂氯化钆 (GC)对阿霉素 (DOX)诱导亚铁血红素氧化酶 1(HO 1)表达的影响。方法 7~ 8周龄Wistar雄性大鼠 ,分为 3组 ,每组 6只 ,分别为正常组 (N) :不做任何处理 ;对照组 (DOX) :尾静脉注入阿霉素 (10mg·kg- 1 ) ;实验组 (GC +DOX) :在注射阿霉素前 1、2天腹腔分别注射 0 2 %的氯化钆溶液 (8mg·kg- 1 )。注射阿霉素后 2 4h ,3组大鼠主动脉采血检测ET 1和TNF α含量 ,切取部分肝脏用蛋白电泳的方法检测HO 1蛋白表达。结果 正常组肝脏组织HO 1表达量很小 ,对照组HO 1表达是正常组的 34倍 ,实验组HO 1比对照组下降了 4 2 %。实验组和对照组血清ET 1和TNF α显著高于正常组 (P <0 0 1) ,实验组血清ET 1和TNF α比对照组略高 (P >0 0 5 )。结论 枯否氏细胞功能抑制剂氯化钆可以抑制阿霉素诱导的HO 1表达 ,枯否氏细胞是肝细胞表达HO
Objective To study the Effect of Gadolinium Chloride on the Induction of Heme Oxygenase 1 Protein by Doxorubicin in rat liver.Methods 7~8 weeks Wistar rats were divided into three groups:Normal group(N):no treatment, Doxorubicin group(DOX):injected Doxorubicin(10 mg·kg -1 ) by teil vein and Gadolinium Chloride plus Doxorubicin group(GC+DOX):Gadolinium Chloride(8 mg·kg -1 ) was injected one and two day before the injection of Doxorubicin. Twenty four hours later After the injection of Doxorubicin, serum ET 1 and TNF α were determined and the expression of HO 1 in rat liver was detected by westernblot.Results 24 hour after the injection of Doxorubicin, the amount of HO 1 in DOX group was 34 times as much as Normal group, and after the treatment of Gadolinium Chloride, the expression of HO 1 in GC+DOX group decreased 42% compared with that of DOX group. The level of serum ET 1 and TNF α increased significantly in the DOX group and GC+DOX compared with the Normal group( P <0 01).Conclusion Gadolinium Chloride can inhibit the expression of HO 1 induced by Doxorubicin in liver rat and the expression of HO 1 depends on the function of Kupffer cell.
