摘要
目的研究氟化物与αD3联合用药对去势大鼠股骨、颌骨力学性质的影响。方法选3月龄SD雌性大鼠分为假手术组、去势组、用药组于去势后6周,用一氟磷酸谷酰胺(0.45 mgF-+13.56 mgCa2+/kg·d)与αD3(αD30.1μg/kg·d)灌胃,第18周末结束实验。测血生化指标并作三点弯曲骨生物力学测量。结果去势后大鼠体重增长迅速,较假手术组比较差异有显著性(P <0.05);去势后雌激素下降明显,与假手术组比较差异有显著性(P <0.05)。去势组碱性磷酸酶水平较假手术组升高(P<0.05),一氟磷酸谷酰胺与αD3联合用药组碱性磷酸酶水平接近假手术组;去势组股骨生物力学指标较假手术组没有明显变化,但氟化物与αD3联合用药组力学指标较假手术组有较大提高。结论一氟磷酸谷酰胺组与去势组相比并不降低生物力学指标,但与αD3联合用药可促进股骨力学强度的提高。
Objective: To investigate the effects of monofluorphosphate and αD3 on the mandibule and femur in ovariectomized rats by three-point bending test. Methods: Sprague-Dawley rats of 90 day's age were randomly divided into the following groups and treated for 12 weeks: sham, ovx and treated groups. Animals of treated groups were received monofluorphosphate for 6 days/week at a dose of 0.45 mgF- + 13.56 mg Ca2+/(kg·d) body weight, by gastric feeding at the beginning of seven weeks after ovariectomy. They were also treated with αD3 at a dose of 0.1 μg/(kg·d) body weight. The blood samples were collected for determination of biochemistry parameters. The left mandibule and fermur were preserved for three-point bending test. Results: Compared with sham group, ovariectomy was followed by an increase in body weight (P <0.05). The estradiol was obviously higher in the sham group than that in the OVX group (P <0.05). The alkaline phosphatase was significantly greater in OVX group compared with the sham group (P <0.05), however, the alkaline phosphatase in MFP+αD3 group was similer to that of sham group. There was no difference in biomechanical index between OVX and sham, but the biomechanical index of MFP+αD3 was better than that of the sham and OVX as well as MFP groups, respectively. Conclusions: The combination of MFP with αD3 increases the indices of biomechanical strength over MFP treated alone.
