摘要
目的研究TNF-αmRNA在实验性兔急性心肌梗死(AMI)心脏不同部位表达的特点。方法结扎兔前降支近端,用原位杂交的方法检测TNF-αmRNA在梗死区、梗死周边区及梗死区对侧左室、左心房、右室和右房的表达。结果在整个观察期内可在梗死区、梗死周边区梗死、梗死区对侧左室及左心房检测出TNF-αmRNA表达,在右室和右房无表达。结论AMI早期兔心脏的梗死区、梗死周边区梗死、梗死区对侧左室及左心房存在TNF-αmRNA表达;左室及左房非梗死区TNF-αmRNA表达的机制可能与心肌梗死后左室和左房壁张力增高有关。
Objective: To determine spatial distribution of tissue expression of TNF-αin an experimental model of acute myocardial infarction (AMI) in rabbits. Methods: Rabbits were randomized to proximal left anterior descending ligation. On days of 1, 7 and 14 after ligation, expressions of TNF-αmRNA were examined by in situ hybridization from the infarct zone, peri-infarct zone, contralateral noninfarct zone of left ventricle, and left atrium, right ventricle and right atrium. Results: There was increased TNF-αmRNA expression throughout the observing periods in the infarct zone, peri-infarct zone, contralateral noninfarct zone of left ventricle and left atrium, but not in the right ventricle an right atrium. Conclusions: The TNF-αmRNA expression is presented early in the rabbit model of AMI. The expression is not only confined strictly to the infarct either peri-infarct zone, but also is presented in the contralateral noninfarct zone of left ventricle and left atrium. The elevated wall stress in left ventrecle and lefet atrium after AMI may be the cause of the TNF-αmRNA expression in the myocardium of the contralateral noninfarct zone of left ventricle and left atrium.
