摘要
利用钙离子选择电极研究了La3+、Cu2+、Zn2+、Cd2+、Mg2+和Al3+等金属离子与拟南芥钙调素(CaM)的竞争结合作用。结果表明:La3+、Cu2+、Zn2+、Cd2+与钙调素的竞争结合能力较强,Al3+和Mg2+的竞争能力较弱。竞争实验的结果从分子水平上证明了金属离子可取代Ca2+4·CaM中的Ca2+与钙调素产生竞争性结合,揭示了这些金属离子生物效应可能的分子机制。
The effects of metal ions La3+,Cu2+,Zn2+,Cd2+,Mg2+and Al3+ on the substrate-binding ability of cauliflower calmodulin have been studied by Ca2+ selective electrode .The competition binding abilities of La2+,Cu2+,Zn2+,Cd2+ to CaM are higher than that of Al3+ and Mg2+.The results comfirmed that metal ions can substrate Ca2+ in Ca42+·CaM system and bind to calmodulin competively,meanwile,the experiment has disclosed the molecular mechanism of the biological effects of La3+, Cu2+, Zn2+,Cd2+,Al3+and Mg2+ at molecular level.
