去甲肾上腺素、牛血清白蛋白诱发血瘀证动物模型的研究

A study on the Experimental Model of Xueyu Syndrome in Rabbits Induced by Nor-Adrenaline and BSA

分别用去甲肾上腺素(NE)、牛血清白蛋白(BSA)、NE+BSA诱发免血瘀证模型,结果三组造型后所检测指标均有不同程度的改变,尤以NE+BSA组改变最为明显。其中,NE+BSA组与正常对照组比较,其高切率全血粘度、低切率全血粘度、血浆粘度、全血还原粘度以及血浆TXB_2含量、红细胞IC花环率均明显升高;血小板1min聚集率与血小板最大聚集率及红细胞聚集均明显增加;血浆6—Keto—PGF_(1α)含量、红细胞C_(3h)受体花环率均明显降低;肠系膜微循环血流流速减慢,以粒流、粒缓流为主。以上各改变与正常对照组比较差别均有显著或非常显著性意义(P<0.05或P<0.01)。提示NE与BSA合用可使造型兔产生高浓、高粘、高聚、高凝的血瘀证病理状态,与血瘀证的临床改变相似,也与血瘀证的中医病理机制相符,故可认为此血瘀证模型为一较稳定、理想的慢性血瘀证模型,可用于血瘀证实质的探讨及活血化瘀方药作用机理的研究。

In this study, an attempt was made to conduct a Xueyu-syndrome model in New Zealand white rabbits induced by injection of nor-adrenaline(NE), bovine serum albumin(BSA), or combination of NE and BSA respectively. Compared with the control group injected with normal saline, and with each other, the results showed that the observed data obtained from the NE-BSA group were markedly different from those in other groups. The findings in NE-BSA group were as follows: 1) high shear rate, low shear rate, plasma viscosity and reduced viscosity in hemorheology were significantly higher; 2) erythrocyte electrophoresis time was markedly longer; 3) platelet aggregation at 1 minute and maximum rate increased significantly respectively; 4) plasma level of TXB_2 greatly increased while plasma level of 6-Keto-PGF_(1α) greatly decreased; 5) RBC C_(3b) rosette rate was lower and RBC immunocomplex rosette rate was significantly higher; 6) in the observation of mesenteric microcirculation, the blood flow decreased with characteristics of grain-line stream and grain stream. The results may be explained by the injure process induced by NE-BSA. Furthermore this pathological state is closely similar to clinical condition of patients with Xueyu syndrome. It is also suggested that the model induced by NE-BSA is more practical in terms of its correspondence with clinical evidence than that induced by NE or BSA alone.
