摘要
目的 研究D3S175 4的遗传多态性及法医学应用价值。方法 应用聚合酶链式反应 (PCR)、聚丙烯酰胺凝胶电泳及银染显带技术对温州地区汉族 184名无关个体的D3S175 4位点进行分型 ,并检验D3S175 4基因型频率分布是否符合Hardy -Weinberg平衡 ,计算法医学常用的各种概率。结果 D3S175 4位点检出 8个等位基因 ,19种基因型。基因型频率的分布符合Hardy -Weinberg平衡 (χ2 =10 .1,P >0 .0 5 ,df=11) ,观测杂合度h0为 75 0± 1 37% ,偶合机率为 0 0 99,亲子关系指数为 2 0 ,个体识别率为 0 90 1,期望排除率为 0 5 10 ,多态信息含量为 0 73,不同人群基因频率分布存在一定的差异。结论 所得到的等位基因频率数据可为温州汉族人群法医个体识别 ,亲子鉴定及遗传学研究提供更多依据。
Objective: To investigate polymorphism of STR locus D3S1754 and its availability in forensic science. Methods: 184 unrelative individuals of Han population living in Wenzhou were examined using PCR followed by PAGE and silver stains, and the examination D3S1754 locus genotype frequency distribute whether to match Hardy-Weinberg or not equilibrium,compute in common use and every kind of rate of forensic science. Results: 8 alleles and 17 genotypes of D3S1754 were observed among the 140 unrelated individuals. The genotype frequency match the Hardy-Weinberg equilibrium(χ 2=10.1,P>0.05,df=11), The observed heterozygosity (h0),Matching Probability, Typical Paternity Index, Power of Exclusion, Polymorphism Information Content(PIC) and Power of Discrimination(DP) were 75.0±1.37%,0.099,2.0,0.510,0.73 and 0.901 respectively. Conchision: The locus in this study roas useful markor for individual indentification and paternity test and for genetics puroses.
