干扰素治疗病理性瘢痕的实验研究

摘要 目的 :研究体外干扰素 (INF)对瘢痕组织成纤维细胞 (FB)中转化生长因子 β1(TGF β1)、血小板衍生生长因子BB(PDGF BB)、基质金属蛋白酶 (MMP 1)以及糖基转移酶 (ppGalNAc T2 )的mRNA表达的影响 ,以求阐明干扰素对病理性瘢痕的作用机理。方法 :组织块法培养瘢痕组织FB ,传代后分成 3组①对照组 :生理盐水处理 ;②低浓度组 :10 0U/mlINF α2b处理 ;③高浓度组 :10 0 0 0U/mlINF α2b处理 ;RT PCR检测TGF β1、MMP 1、PDGF BB及ppGalNAc T2表达。结果 :体外培养瘢痕FB经 10 0U/ml及 10 0 0 0U/ml干扰素α 2b处理后 ,RT PCR检测TGF β1、PDGF BB及pp GalNAc T2表达降低 ,MMP 1表达升高 ,呈显著差异 ,且具有浓度依赖性。结论 :干扰素α2b对瘢痕FB具有抑制作用 ,其机理可能与多种细胞因子有关 ,如TGF β1、PDGF BB。 Objective: To research the effect of interferon on TGF-β_1,MMP-1,PDGF-BB and ppGalNAc-T_2in FB of pathological scars,and the mechanism of interferon on pathological scars.Methods: The FB of pathological scars were cultured by the method of tissue culture and were divided randomly in three groups: ①control(0.9%sodium chloride);②low concentration (100 U/ml INF-α2b);③ high concentration ( 10,000 U/ml INF-α2b). Then total groups were analysed the expression of TGF-β_1,MMP-1,PDGF-BB,and ppGalNAc-T_2with RT-PCR.Results: After the effect of 10 U/ml INF-α2b and 10,000 U/ml INF-α2b on cultured fibroblast of pathological scars,the expression of TGF-β_1,PDGF-BB and ppGalNAc-T_2were lower than that of controlled group and the expression of MMP-1 was higher than that of controlled group. The result was significantly different and was related with the concentration of INF-α2b.Conclusion: The reason of the good effect of INF-α2b on inhibiting fibroblast of pathological scars may be related with some kinds of cellula factor,such as TGF-β_1,PDGF-BB,bFGF,et al.
