摘要
目的 :与以往术前 2 1天进行胸腺修饰常规方法不同 ,在手术当天进行胸腺内注射抗原 ,诱导大鼠同种心脏移植免疫耐受。方法 :在大鼠腹腔异位心脏移植模型中 ,实验组在手术当天将供体SD大鼠脾细胞注射到受体Wistar大鼠胸腺 ,并以FK5 0 6短程处理受体鼠。单纯药物对照组在围手术期使用FK5 0 6但不进行胸腺内注射供体脾细胞 ;经典方法诱导组 ,腹腔注射抗淋巴细胞血清ALS( 1天 ) +胸腺内注射供体脾细胞 ( 2 1天 ) ;无处理组 ,单纯将SD大鼠心脏移植到Wistar大鼠腹腔。观察供心存活天数、病理改变及混合淋巴细胞反应等指标的变化。结果 :实验组供心存活时间较对照组明显延长 ,而与经典诱导组比较无统计学差异。实验组和经典诱导组的供受体脾细胞混合淋巴细胞培养各组增殖反应均较无处理组降低 ,而两者之间无差异。结论 :心脏移植手术当日胸腺内注射供体同种抗原 ,同时给予短程免疫抑制剂与术前 2 1天胸腺注射的经典诱导组同样能诱导宿主对移植物的低反应状态 (免疫耐受或免疫抑制 )
Objective:To induce tolerance to cardiac allografts by combining intrathymic inoculation with allogeneic antigen at the day of transplantation which is different from the conventional measure. Methods:2.5 ×10 7allogeneic spleen cells of SD rats were intrathymically injected into Wistar rats at the time of cardiac allografting from SD rats with short-course therapy of FK506 in this experimental group. In control group, allogeneic spleen cells (Sc) of SD rats were intrathymically injected into Wistar rats at the time of cardiac allografting from SD rats without short-course therapy of FK506. Survival time of cardiac allografts, the histological changes in SD cardiac allografts, the mixed lymphocyte reaction (MLR) between donors and recipients were investigated.Results:The MST of cardiac allografts in experiment group is longer than that in control group. The results of MLR showed that spleen T cells harvested from tolerant Wistar rats displayed hyporesponsiveness against naive SD spleen cells, and the proliferous responses of no pretreatment group were greater than those in the tolerant rats. Conclusion:Tolerance was obtained by intrathymical injection of donor spleen cells on the day of transplantation and short course of usage of post-grafting.
