摘要
目的 :通过研究体外培养的海马神经元被戊四氮活化后的产物与癫痫的关系 ,探讨惊厥发作前癫痫形成的机制。方法 :将神经元活化产物注入大鼠的侧脑室 ,观察大鼠行为、脑电图及GABA(α1)受体免疫组织化学反应的改变。结果 :实验组注射神经元活化产物后 10~ 30min大部分大鼠出现 2~ 3级惊厥反应 ,EEG呈短程、中高幅尖波、尖慢复合波 ;对照组行为及EEG未见异常 ,实验组GABA(α1)受体免疫组化反应呈阳性的细胞明显少于对照组。结论 :神经元活化产物具有致痫作用 ,可通过抑制GABA(α1)
Objective: To investigate the effect of the activated hippocampal neuronal production and try to found out the epileptogenesis before seizure. Methods: The activated hippocampal neuronal products were injected into rat’s lateral ventricle and the changes of rat’s behavior, EEG, immunohistochemical reaction of GABA(α 1)R were observed. Results: During 10-30 min, after the administration of PTZ activated hippocampal neuronal production in lateral ventricle, Ⅰ-Ⅲ class epileptic behavior was found in the experimental group, meanwhile corresponding change in electroencephalogram occurred, which showed moderate amplitude sharp waves, while no similar change was found in control group. Compared with the control group, the expression of GABA(α 1)R in immunohistochemical reaction positive cells was decreased obviously. Conclusion: Activated hippocampal neuronal production could induce seizure. One of epileptogensis might be related to the expression change of GABA(α 1)R by the effect of NF κВ.
