摘要
目的 :探讨体外分离、培养及鉴定前软骨干细胞 (PCSC) ,为相关的实验研究奠定基础。方法 :采用酶消化法从新生大鼠干骺端中收集细胞 ,用免疫磁珠法分选出前软骨干细胞 ,用DMEM F12培养基进行体外培养 ,用免疫组化方法鉴定 ,对分离出的前软骨干细胞进行相关分子生物学研究。结果 :从新生大鼠干骺端中成功培养出前软骨干细胞 ,培养条件下贴壁生长 ,成团块状 ,细胞表面有FGFR 3的标记物 ;细胞生长迅速。结论
Objective: To isolate, cultivate and identify the precartilage stem cells from neonatal rat in vitro for the further related research. Methods: The cells from the neonatal rat’s metaphysics were digested by enzyme; The precartilage stem cells were isolated by immuniomagnetic beads coasted with the second antibody; DMEM F12 medium was adapted to culture the cells; The cells were identified by immunocytochemistry. The precartilage stem cells were studied by molecular biology technology. Results: Precartilage stem cells from neonatal rat have been successfully cultured. They formed typical growth in mass. There was FGFR 3 on the surface of the precartilage stem cells. Conclusion: Precartilage stem cells could be cultured from neonatal rat in vitro. They may be potential source for transplantation in treating defection of bone and cartilage.
