不同处理的NS_1骨髓瘤细胞微小池分隔培养对树突状细胞生长和活性的影响

摘要 目的 :观察不同方法处理灭活的骨髓瘤NS1细胞对树突状细胞 (DC)生长和活性的影响。方法 :采用细胞冻融、紫外线照射、次黄嘌呤 氨基嘌呤 胸腺嘧啶核苷 (HAT)杀死肿瘤细胞法处理NS1细胞 ,将处理过的NS1细胞与DC细胞分别于直径 3μm和直径 8μm微小池内同池培养 ,并以活NS1细胞和空白微小池DC孔为对照 ;观察DC数目和形态改变 ;检测各组DC激活的体外混合淋巴细胞杀伤活性 ,观察各组DC对NS1骨髓瘤动物移植性肿瘤的抑瘤效果。结果 :与活NS1同一孔池培养的DC数目减少 ,形态改变 ,不能诱导太强的细胞活性T淋巴细胞 (CTL)作用和宿主体内免疫系统的抗肿瘤作用。而不同处理灭活NS1脉冲致敏的DC增殖旺盛 ,形态典型 ,均诱导出较强的CTL作用和宿主体内免疫系统的抗肿瘤作用。结论 :在肿瘤免疫治疗中 ,脉冲致敏DC一定要用灭活的肿瘤细胞作为抗原刺激物 ,才能有效地完成抗原递呈功能。 Aim: To observe effects of NS 1 myeloma cells treated by different methods on growth and activity of dendritic cells separately cultured in same well. Methods: NS 1 myeloma cells within logarithmic growth were separately devitalized by freeze-thaw, ultraviolet ray shining, and HAT, then cultured with DC in the same well and separated by 2 microwells with different size aperture (3 μm and 8 μm). DC cultured only and with vitalized NS 1 were the controls,respectively.7 days later,the numbers and shapes of DC, the cytotoxic effect of lymphocytes activated by DC, and the inhibition to NS 1-transplanted tumor of DC were were observed.Results: The number decrease and shape changes of DC cultured with vitalized NS 1 in same well were observed. And these DC could not induce more powerful effects of CTL and antitumor effect in host body. As to DC cultured with the NS 1 treated by other different methods,their growth was vigorous, shapes were typical and these DC could induce relatively powerful effects of CTL and antitumor effect in host body after sensitization. Conclusion: DC with pulse sensitization should use deactivated tumor cells as antigen stimulator substance, then, effectively perform antigen-presenting function in immune treatment of antitumor.
