摘要
用经典酚醇法、盐析法、碘化钾法、煮沸法提取外周血单个核细胞DNA ,比较这四种方法所获得的DNA模板对PCR检测肿瘤坏死因子 (TNF)的影响 ,结果显示除了煮沸法 ,其余三种方法提取的DNA均有较高的纯度和浓度 ,其中以碘化钾法操作最为简便可靠 ,适合临床推广应用。
DNA of peripheral blood mononuclear cell(PBMC) was extracted by four different kinds of methods, which were classical hydroxyketone assay, salting out assay, potassium iodide assay, and boiling assay .The obtained DNA were used as template to detect tumor necrosis factor(TNF) by polymerase chain reaction (PCR) and to approach the effect of different template on PCR. The results showed that among the four methods, the potassium iodide technique was the simplest and the most suitable method in routine extraction of PBMC DNA.
