人参银杏复方制剂对缺氧复氧后大鼠海马CA1区Fos表达的影响 被引量:3

摘要 目的:观察人参银杏复方制剂对缺氧24h复氧后不同时段Fos表达时程变化的影响,探讨人参银杏复方制剂抗缺氧复氧性脑损伤机制。方法:35只清洁级SD大鼠随机分为常氧对照组,缺氧复氧实验组和人参银杏复方制剂给药组。应用低压氧舱仿海拔8000m高空缺氧模型,采用Nissl染色、Fos免疫组化方法结合图像分析技术,观察预防性应用人参银杏复方制剂对缺氧24h复氧0,24,72h不同时段海马CA1区锥体细胞层神经元形态及Fos免疫反应阳性神经元的影响。结果:全脑缺氧24h复氧72h后,海马CA1区锥体细胞层神经元数量锐减,Fos免疫反应阳性神经元数量在缺氧24h复氧0h时为(9.3±3.8)个,较常氧对照组(16.6±4.8)个减少,差异有显著性意义(P<0.05),复氧24h时显著减少甚至消失为(2.0±1.8)个,复氧72h为(13.7±5.1)个,时接近常氧对照组水平差异无显著性意义(P>0.05)。预防性应用人参银杏复方制剂后缺氧24h复氧72h时海马CA1区锥体细胞层神经元数量未见明显减少,在缺氧24h复氧0,24,72h时段CA1区Fos免疫反应阳性神经元数量分别较缺氧复氧实验组相应时段增加,以复氧0h时增加最为显著,为(39.0±5.8)个,与常氧对照组比较差异无显著性意义(P>0.05)。结论:人参银杏复方制剂对缺氧复氧后海马CA1区神经元具有保护作用。 AIM:To observe the effect of renshen ginkgo extract compound on the changes of Fos expression at different stage after hypoxia and 24-hour reoxygenation,and to explore the mechanism of renshen ginkgo extract compound against hypoxia-reoxygenation brain injury. METHODS:Thirty-five SD rats of cleaning grade were randomly divided into normal oxygen group(control group),hypoxia-reoxygenation group(experimental group)and renshen ginkgo extract compound treatment group(administration group). The hypoxia models at an 8 000 meter high level were established in a hypobaric oxygen chamber.Nissl staining,Fos immunohistochemistry and image analysis technique were used to observe the effect of preventive application of renshen ginkgo extract compound on the morphology of pyramid neurons and the number of Fos-like immunoreactive neurons in CA1 area of hippocampus at different stages after hypoxia for 24 hours followed by reoxygenation for 0,24 and 72 hours. RESULTS:After hypoxia for 24 hours followed by reoxygenation for 72 hours,the number of pyramid neurons in CA1 area was decreased sharply,the number of Fos-like immunoreactive neurons after hypoxia followed by reoxygenation for 0 hour(9.3±3.8)was significantly less than that in the control group(16.6±4.8)(P< 0.05)and even disappeared until reoxygenation for 24 hours(2.0±1.8),but that of reoxygenation for 72 hours(13.7±5.1)was not significantly different from that in the control group(P >0.05).In administration group, the number of pyramid neurons in CA1 area of hippocampus did not change obviously after hypoxia for 24 hours followed by reoxygenation for 72 hours, and the number of Fos-like immunoreactive neurons in CA1 area of hippocampus after reoxygenation for 0,24 and 72 hours were increased significantly than that in the experimental group in the corresponding period with the most obviously at 0 hour of reoxygenation(39.0±5.8),which was insignificant as compared with that in the control gorup(P >0.05).CONCLUSION:Renshen ginkgo extract compound plays a role in protecting neurons in CA1 area of hippocampus after hypoxia-reoxygenation, and its mechanism may be related with the enhancement of the Fos expression in CA1 area of hippocampus after hypoxia-reoxygenation.
