背景:人类对脑出血的研究常采用大鼠的脑出血模型,但脑出血模型中缺乏一些重要指标描述。目的:为脑出血的发病机制、治疗等研究提供有效可靠的疾病动物模型及相关指标。设计:随机对照实验研究。地点和对象:成年SD大鼠50只,均为雄性,实验在四川大学基础与法医学院人体解剖学教研室完成。干预:SD大鼠50只随机分为自体血注入组(20只)和胶原酶注入组(20只),在大鼠尾状核分别注入自体血50μL和胶原酶5.001nkat,建立两种不同的脑出血模型,术后3d自体血注入组大鼠死亡5只,胶原酶注入组死亡共6只。主要观察指标:两组大鼠神经功能学评分,病理变化,血肿大小,免疫组织化学染色结果。结果:血肿大小犤自体血注入组为(10.65±3.12)μL、胶原酶注入组为(40.26±1.45)μL犦,两者比较差异有统计学意义(t=33.102,P<0.001)、阳性小胶质犤自体血注入组为(32.6±3.6)个、胶原酶注入组为(8.8±2.4)个犦,两者比较差异有统计学意义(t=20.885,P<0.001)。结论:由于两种方法建立的脑出血模型各种指标不同,因此可以根据不同的研究目的采用不同的动物模型。
BACKGROUND:The models of intracerebral hemorrhage in rats are often used to study the intracerebral hemorrhage in human beings,but some of the important indexes descriptions are not available in those models. OBJECTIVE:To provide effective and reliable animal models and some related data for the research of pathogenesis and treatment of intracerebral hemorrhage.DESIGN:A randomly controlled experimental study.SETTING and MATERIALS:Fifty adult male SD rats were selected.Trials were conducted in the Department of Human Anatomy of the College of Preclinical and Forensic Medicine of Sichuan University.INTERVENTIONS:Fifty SD rats were randomly divided into two groups: autoblood-injected group and collagenase-injected group.50 μL of autologous blood and 5.001 nkat of collagenase were respectively injected into the caudate nucleus in rats to make two different kinds of animal models of intracerebral hemorrhage.Three days after the operation,5 rats of the autoblood-injected group died and 6 rats of the collagenase-injected group died.MAIN OUTCOME MEASURES:Neurologic function rating,pathological changes,volume of hematoma and immunohistochemistry of these two groups.RESULTS:There was a statistical significance in the volume of hematoma[autoblood-injected group:(10.65±3.12) μL;collagenase-injected group:(40.26±1.45) μL] between these two groups(t=33.102,P< 0.001). There was a statistical significance in the number of positive microglias cells[autoblood-injected group:(32.6±3.6);collagenase-injected group:(8.8±2.4)] between these two groups(t=20.885,P< 0.001).CONCLUSION:Because of the differences of the indexes between these two animal models,we can adopt different models in research according to different purposes of studies.
