摘要
目的:以往实验显示补益生肌法有促进创面修复和减少瘢痕形成的作用,为了解其作用机制,在创面修复早、中期观察该法对实验性创面新生肉芽组织中I,III型胶原和胶原金属蛋白酶-1(matrixmetalloproteinase-1,MMP-1)mRNA表达的影响。方法:32只大鼠创面造模后随机分为喂药3,10d两个时段,每个时段分为生理盐水模型组、补益生肌方组,每组各8只。采用Northernblot杂交法对大鼠创面肉芽组织中胶原MMP-1和I,III型胶原mRNA在创面修复早期(3d)和中期(10d)表达进行检测。结果:显示在创面修复3d时,补益生肌组的MMP-1的mRNA表达弱于模型组,I,III型胶原的mRNA表达与模型组相似。创面修复10d时,补益生肌组MMP-1mRNA表达弱于模型组,I,III型胶原的mRNA表达强于模型组。创面修复初期(3d)和中期10d)对比:补益生肌组MMP-1的(mRNA表达降低;I型胶原mRNA表达略降;III型胶原无明显变化。模型组MMP-1的mRNA表达无明显变化;I型胶原mRNA表达明显下降;III型胶原mRNA表达明显下降。结论:补益生肌法对实验性创面新生肉芽组织中I,III型胶原和MMP-1mRNA的表达的影响可能是在创面愈合的过程中,通过抑制MMP-1的分泌,从而使创面I,III型胶原的分泌增加,起到促进创面愈合的作用。
AIM:Previous experiments showed that the therapy for invigoration and promoting tissue regeneration has the effect of prompting wound healing and reducing scar forming.In order to know the mechanism,we observed the effect of this method on the expressions of mRNA of Ⅰ and Ⅲ type procollagens and matrix metalloproteinase 1(MMP 1) in experimental wound granulation tissue. METHODS:Thirty two rats were randomly divided into two phases after their wound made into models: administration for 3 days and administration for 10 days.The rats in each phase were divided into saline model group and therapy for invigoration and promoting tissue regeneration group,8 in each group.The expressions of mRNA of I and Ⅲ type procollagens and MMP 1 of wound granulation tissues in rats in the early phase(3 days) and the middle phase(10 days) of wound healing were detected with the Northern blot hybridization method. RESULTS: On day 3 of wound healing, the mRNA expression of MMP 1 in the therapy for invigoration and promoting tissue regeneration group was lower than that in the model group,and the mRNA expressions of I and Ⅲ type procollagens were similar with those in the model group.On day 10 of wound healing,the mRNA expression of MMP 1 in the therapy for invigoration and promoting tissue regeneration group was lower than that in the model group. The comparison between the early phase(3 days) and the middle phase(10 days) showed that the mRNA expression of I and Ⅲ type procollagen in the therapy for invigoration and promoting tissue regeneration group decreased, the mRNA expression of I type procollagen decreased slightly,and Ⅲ type procollagen had no obvious changes;The mRNA expression of MMP 1 in the model group had no obvious changes,and the mRNA expressions of I and III type procollagens decreased obviously. CONCLUSION:The effect of therapy for invigoration and promoting tissue regeneration on the mRNA expressions of I and Ⅲ type procollagens and MMP 1 in experimental granulation tissue maybe increases the excretion of I and Ⅲ type procollagens and promotes the wound healing by inhibiting the excretion of MMP 1 in the course of wound healing.
