背景:淫羊藿是中医治疗骨质疏+松症常用中草药中使用频率最高者之一,但对其抗骨质疏松有效成分的研究尚不多见。目的:研究淫羊藿总黄酮提取物(TFE)的抗骨质疏松活性。设计:完全随机,前后和组间对照实验研究。地点和对象:在解放军兰州军区兰州总医院动物实验科完成,清洁级SD大鼠48只,由甘肃省医学科学研究院实验动物中心提供。干预:48只大鼠分为6组,行双侧卵巢切除术OVX或假手术。3组行()OVX者分别以50,100和200mg/(kg·d)的剂量灌服TFE,1组行OVX者灌服1.5mg/(kg·d)的尼尔雌醇作为阳性对照。3个月后处死所有大鼠。主要观察指标:术后大鼠血尿生化指标、骨组织形态学、骨密度和股骨力学强度检查。结果:OVX导致骨微结构退化、骨量丢失和股骨力学强度下降。OVX组血清ALP为219.41±34.52)U/L,显著高于正常对照组;TFE200和(尼尔雌醇组分别为(164.32±37.96)和(150.30±44.83)U/L,显著低于OVX组;血清OC浓度变化与ALP相似。尿Ca/Cr,P/Cr和DPD/Cr水平OVX组均显著高于正常对照组,尼尔雌醇组各项指标均显著低于OVX组t=4.17~4.50,P<0.05)。OVX组股骨骨密度显著低于正常(对照组,TFE200和尼尔雌醇组显著高于OVX组。L2~L4腰椎变化与股骨相似。灌服TFE抑制或减弱了OVX的作用,尤其200mg/(kg·d)的疗效与尼尔雌醇相当。
BACKGROUND:Epimedium herb has been one of the most frequently used herbs in treating osteoporosis in traditional Chinese medicine. However,its anti osteoporotic constituents have rarely been studied. OBJECTIVE:To investigate whether the total flavonoid extract(TFE)of Epimedium sagittatum has an anti osteoporosis activity. DESIGN:A completely randomized,auto controlled and mutual controlled study was used. SETTING and PARTICIPANTS:Forty eight adult female SD rats provided by the Department of Animal Experiment of Lanzhou General Hospital Area Command of Chinese PLA.were used and the experiment was completed in the Department of Animal Experiment of Lanzhou General Hospital of Lanzhou Military Area Command of Chinese PLA. INTERVENTIONS:Forty eight rats were divided into 6 groups and underwent bilateral ovariectomy(OVX)or sham operation.TFE were orally administered at 50,100 and 200 mg/kg per day separately.Nilestriol(E2) was given at 1.5 mg/kg per day and used as positive control.All rats were sacrificed after three months. MAIN OUTCOME MEASURES:The biochemical and bone histomorphometric parameters,bone mineral density(BMD) and breaking strength of femurs were measured. RESULTS:OVX caused bone micro architectural deterioration and significant decreases in both of the bone mass and mechanical strength of femur.The serum alkaline phosphatase(ALP)[(219.41±34.52)U/L] in OVX was obviously higher than that in normal control group;TFE200 and E2,(164.32±37.96)and(150.30±44.83)U/L respectively,were obviously lower than those of the OVX group;the level of serum isteocalcin(OC) was similar to OVX group.Urine Ca/Cr,P/Crand DPD/Cr was obviously higher than that of the normal control group;All the markers of E2 group were lower than those of OVX group(t=4.17-4.50,P< 0.05).The BMD of femur in OVX was obviously lower than in normal control group.Each marker of TFE200 and E2 were obviously higher than those of OVX group.Change in Lumbar vertebrae of L2-4 was similar to that in femur.TFE inhibited or attenuated the effect of OVX,especially at 200 mg/kg per day,which exerted almost the same preventive effect as nyestriol without exhibition of uterotrophic activity. CONCLUSION:TFE has an anti osteoporotic activity.
