免疫磁珠法分离和纯化重症肌无力患者骨髓CD34^+造血干细胞

摘要 目的 分离和纯化重症肌无力 ( MG)患者骨髓 CD3 4+造血干细胞。方法 采用免疫磁珠法 ( Mini MACS)分离和纯化 CD3 4+ 细胞 ,用流式细胞仪对其进行评估并检测纯化后细胞活力。结果 骨髓分离纯化前、后CD3 4+细胞的百分率分别为 ( 2 .65± 0 .65 ) %、( 85 .3 7± 7.47) % ( P<0 .0 0 1 ) ;CD3 4+细胞分离纯化前、后细胞活力未受影响 ( P >0 .0 5 )。结论 Mini MACS可有效地分选 MG患者 CD3 4+骨髓造血干细胞 ,且细胞活力不受影响。 Objective Isolation and purification of the bone marrow CD34 +hematopoieticstem cells were undergone to study the biological characteristics of CD34 + hematopoietic stem cell and its clinical transplantation in myasthenia gravis (MG) patients. Methods The bone marrow CD34 + cells were isolated and purified by means of imunomagnetie beads on MiniMACS device. The percentage of CD34 + cells was measured by flow cytometry before and after MiniMACS. Results The percentage of CD34 + cells before and after the purification were (2.65±0.65)% and (85.37±7.47)% resprectively,and the cell ability was not affected by purification procedure. Conclusions MiniMACS can sort out effectively the bone marrow CD34+ cells and does not affect the cell vitality.
