Synthesis and characterization of immunogen medroxyprogesterone acetate residues in edible tissue

摘要 采用碳二亚胺法和混合酸酐法将MPA与牛血清白蛋白偶联,用紫外分光光度法估测其交联率分别为14和20,从SDS聚丙烯凝胶电泳条带可看出交联成功。经间接酶联免疫测定,进一步证明甲羟孕酮免疫原合成成功。 Medroxyprogesterone acetate (MPA), as a half-antigen can′t lead to immune reaction and has to be conjugated with a carrier, such as bovine serum albumin (BSA). Two methods were used to prepare the immunogen including the carbodiimides reaction and the mixed anhydride reaction. The conjugates of MPA-BSA were identified by the analysis of spectrophotometry, SDS electrophoresis and animal immunization test.The conjugated ratios were 14 and 20 respectively. The results demonstrated that the synthesis of MPA-BSA were successful.
