从柯萨奇B病毒(CVB)衣壳蛋白VPI的保守区选出一段13肽进行化学合成,经ELISA证买该段多肽是CVB的共同优势抗原表位.以多肽作包被抗原,建立了检测CVBIgM的间接ELISA法,对85例健康人、230例病毒性心肌炎、19例RF阳性血清、40例其它疾病患者进行了检测,阳性率分别为7.1%、41.3%、21.1%、10%.230例心肌炎中有106例用以病毒作包被抗原的间接ELISA进行了CVBIgM检测,阳性率49.1%.两者之间结果有良好的相关性(X^2=41.9,P<O.0001),阳性率无明显差别(X^2=2.45,P>0.05).提示,应用合成肽抗原检测CVBIgM抗体有较好的敏感性和特异性.
A 13 amino acid peptide derived from the conserved regions of CVB coat protein VP1 was synthesized chemically. It was demonstrated that the peptide represented a common immunodomi-nant epitope of CVB coat protein VP1. Indirect ELISA technique for detecting the IgM antibodies to CVB using the synthetic peptide as the coating antigen was established. Eighty-five healthy controls, 230 patients with viral myocarditis, 19 patients with rheumatoid factor, and 40 other patients were tested. The CVBIgM positive rate was 7. 1% , 41. 3% , 21. 1% and 10% , respectively. One hundred and six patients with viral myocarditis were also tested for the CVBIgM by indirect ELISA using CVBl-6 viron as the coating antigen. The positive rate (49. 1%) was similar to that obtained by using the peptide (X2 = 2. 45,P>0. 05). A significant correlation between the indirect ELISA using the peptide as the coating antigen and that using CVBl-6 viron as the coating antigen for the IgM antibodies was found (X2 = 41. 9,P<0. 0001). Indirect ELISA by using the synthetic peptide as the coating antigen was showing sensitively and specifically for detecting IgM antibodies to CVB.
