流感病毒裂解过程中的形态变化规律 被引量:6

摘要 目的 研究流感病毒在裂解剂作用下不同时间的形态变化及对病毒亚单位结构形态上的影响 ,掌握流感病毒在裂解剂作用下的裂解规律。方法 将纯化后的流感病毒抗原按 0 5 %~ 0 6 %比例加入裂解剂TritonN 10 1,分别在 0 5 ,1,2 ,3,3 5 ,4 ,5 ,6h取样 ,用有膜电镜载网粘取样品 ,用 2 %磷钨酸 (PTA)负染 ,电镜下观察。结果 流感病毒裂解可大致分为基质蛋白 (Mprotein ,MP)包膜膨胀阶段 ,神经胺酸酶 (NeuraminidaseNA)、血凝素 (Hemagglu tininHA)、脂质双层 (LipidbilayerLB)脱落阶段 ,基质蛋白包膜破裂核衣壳 (NucleocapsidNP)溢出阶段、基质蛋白溶解阶段。结论 采用震荡措施可使病毒间裂解进程趋于同步。 Objective To realize the transformation shape of influenza virus under the action of split,and the action about the virus substructure shape.To learn split way.Methods To draw split influenza viruse specimen of A 1,A 3 and B subtypes at different time,and observe under electron microscope.The split solvent is Triton N-101.Results The splitting of influenza virus can be divided into four stages.The stage includ M protein(Mp) membrane swell neuraminidase(NA) hemagglutinin(HA) lipid bilayer(LB) break away phase,Mprotein(Mp) membrane disrupt,nucleocapsid(NP) spill and Mprotein dissolved phase.Conclusion The method of vibration makes split better.
