摘要
目的 :探讨Th源细胞因子和CD86在哮喘发病中的作用及CD86mAb治疗哮喘的临床潜在价值。方法 :随机选择哮喘急性发作期患儿 2 8例 ,男 18例 ,女 10例 ,年龄 1岁~ 8 0 8岁 ,平均年龄 (4 83± 1 99)岁 ;对照组 15例 ,男 7例 ,女 8例 ,年龄 3 2 5岁~ 10岁 ,平均年龄 (6 6 3± 1 91)岁。ELISA检测PHA刺激并用小鼠抗人CD86mAb干预PBMC(peripheralbloodmononuclearcell,PBMC)后其培养上清IL 4、IL 13、IFN γ水平。结果 :小鼠抗人CD86mAb(5× 10 - 3μg ml)干预PBMC后 ,正常组和哮喘组IL 4、IL 13和IFN γ水平均降低 ,与相应小鼠IgG对照组相比差异有显著性 ,而哮喘组IL 4和IL 13降低更明显。结论 :CD86mAb的阻断是非特异性的 ,即对Th1和Th2细胞均有一定的抑制作用 ,但对Th2细胞的抑制作用尤甚 ,说明CD86信号传导在哮喘的发生和发展过程中起重要作用。
Objective:To explore the role of T lymphocytes activation co-stimulation pathway in asthma pathogenesis and the ability of therapy asthma with Anti-CD86mAb.Methods:The blood samples were taken from 28 asthma children( including 18 male and 10 female, age 1 year-8.08 years) and 15 normal children( including 7 male and 8 female, age 3.25 years-10 years).ELISA was used to detect the levels of IL-4、IL-13、IFN-γ in culture supernatants of PBMC stimulated with PHA and treated with mouse anti human CD86mAb. Results:①When treated control PBMC with anti-CD86mAb, the level of IL-4 in control group(13.30±4.66 pg/ml) was lower than that of mouse IgG control group (15.20±5.22 pg/ml,P<0.05); the level of IL-13 in control group(677.97±278.11 pg/ml) was lower than that of mouse IgG control group (748.15±318.87 pg/ml, P<0.05); the level of IFN-γ in control group(17.55±8.93 ng/ml) was lower than that of mouse IgG control group (34.87±16.26 ng/ml ,P<0.05);②When treated asthma PBMC with Anti-CD86 mAb, the level of IL-4 in asthma group(24.64±12.17 pg/ml) was lower than that of mouse IgG control group (37.14±17.26 pg/ml,P<0.01); the level of IL-13 in asthma group(836.50±402.54 pg/ml) was lower than that of mouse IgG control group (1257.50±632.32 pg/ml,P<0.01); the level of IFN-γ in asthma group(16.14±6.20 ng/ml) was lower than that of mouse IgG control group (17.75±6.41 ng/ml ,P<0.05).Conclusion:The results showed that CD86 might play an important role in asthma, It seems to be an effective method to treat asthma using anti-CD86 monoclonal antibody.
