摘要
目的:观察神经营养素及受体在脊髓损伤后的表达变化。方法:SD大鼠30只,设正常组、假伤组、脊髓损伤组。Allen's法复制5g×10cm脊髓损伤模型。用免疫组化和免疫电镜方法观察各组神经生长因子(NGF)、脑源性神经营养因子(BDNF)、神经营养素-3(NT-3)及其受体TrkA,TrkB,TrkC表达的变化。结果:正常大鼠脊髓神经营养素及其受体表达较少,损伤后表达明显增加。NGF和BDNF伤后1~7d持续高表达,NT-3、TrkA、TrkB和TrkC伤后1~3d表达较高,与正常组和假伤组相比,差异有显著性意义。结论:神经营养素及受体在急性脊髓损伤中表达增加,起保护性作用。
Objective: To explore the expression of neurotrophins and its receptors in spinal cord injury. Method :Thirty Sprague Dawleys rats were randomly divided into three groups:normal, laminectomy, both laminectomy and traumatic injury. The SCI animal model was made by a modified Allen's weight-drop device(50g·cm)on T_(12). Electron microscope and immunohistochemical method was used to observe the changes of NGF, BDNF, NT-3, and TrkA, TrkB, TrkC in each group. Result: After injury, the neurotrophins and its receptor had some significant changes. Compared with the normal and control groups, the labeling cells of BDNF and NGF were more from 1d to 7d after SCI, those of NT-3, TrkA, TrkB and TrkC were more from 1d to 3d. Conclusion: The expression of neurotrophins and its receptors increase in spinal cord injury, neurotrophins may have some protective effects on acute spinal cord.
