摘要
目的 构建含有针对NDRG2的shRNA的质粒 ,抑制NDRG2在HHCC细胞中的表达。方法 用PCR方法从人基因组DNA中扩增出 336bp的U6 启动子 ,与 2 9bp的NDRG2靶序列的反向重复序列和pSNAV质粒相连。连接后的pSNAVU6 质粒转染入HHCC细胞 ,检测它们对NDRG2表达的影响。结果 pSNAVU6 重组质粒能抑制NDRG2的表达。
Objective To construct a plasmid containing the shRNA of NDRG2 and inhibit the expression of NDRG2 in HHCC cells.Methods A 336 bp DNA fragment of human U_6 shRNA promoter was amplified from human genome DNA by PCR,ligated with a 29 bp DNA fragment of reverse repeated motif of NDRG2 and transformed to plasmid pSNAV. The recombinant pSNAVPU_6 plasmids were transfected into (HHCC) cells, and their effects on NDRG2 expression were studied.Results The recombinant pSNAVPU_6 plasmids inhibited the expression of NDRG2 significantly.Conclusion The shRNA of NDRG2 can inhibit the expression of NDRG2 in HHCC cells.
