Preliminary genetics study of five pedigrees with high incidence of affective disorder

摘要 目的探测儿茶酚氧位甲基转移酶 (COMT)基因、载脂蛋白E(Apo -E)基因及五羟色胺受体 2A( 5 HT2a)基因多态性与情感障碍之关系 ,搜寻中国汉族人情感障碍易患性基因。方法应用聚合酶链反应 (PCR)和限制性片段长度多态性 (RFLP)方法对 5个情感障碍高发家系进行初步研究。结果用改进的传递 /不平衡 (TDT)检测结果表明 ,在 5个情感障碍高发家系中 ,COMT基因与情感障碍相关联 ( P <0 .0 5 ) ,而Apo -E基因和 5 HT2a基因不与情感障碍关联 ( P >0 .0 5 )。结论在我们研究的家系中 ,2 2号染色体和长臂 ( 2 2q11.2 )可能存在情感障碍易患性基因。 Objective To explore the relationship between polymorphism of COMT gene and Apo-E gene and 5-HT 2a gene with affective disorder. Search for a gene predisposing to affective disorder in the Han nationality of Chinese. Methods Five pedigrees with high incidence of affective disorder were preliminary studied by polymerize chain reaction(PCR) and restriction fragment length polymorphism(RFLP) technique. ResultsAnalysis results of statistics of the transmission/disequilibrium test (TDT) handing showed that the COMT gene is association with affective disorder (P<0.05),the Apo-E gene and the 5-HT 2a gene are not association for affective disorder(P>0.05) in the five pedigree with high incidence affective disorder.ConclusionOur study results suggest that maybe a affective disorder liability gene on 22chromosome (22q11.2) in our studied pedigrees.
