摘要
在实验室分离和纯化链霉亲和素的基础上对其性质进行了鉴定.在SDS-PAGE中链霉亲和素只有一条带,单体的分子量为14500Da,链霉亲和素的相对分子量约为58000Da;其等电点为5.9-6.2;结合生物素的活性为15.7U/mg.结果表明自制链霉亲和素为核心链霉亲和素,在纯度、活性等方面达到了理想的效果.
On the basis of previous isolation and purification of Streptavidin(SA) , the authors identify its characteristics. The molecular weight of SA was about 58000 Dalton by SDS-PAGE and its isoelectric point was 5.9-6.2 by IEF-PAGE. The averaged biotin - binding activity was 15.7U/mg. The results showed the SA purified by authors is core Streptavidin, which brings a-bout ideal effect as far as purity and activity is concerned and is thus reliable for immunological application and can be used as the substitute for imported SA.
