高效液相色谱(HPLC)技术鉴定中国南方大豆品种异黄酮主要组分 被引量:12

Determination of Major Isoflavone Components Based on HPLC Technology in Southern Soybean Varieties in China

摘要 采用高效液相色谱 (HPLC)技术检测中国南方六个省份的 2 4 9份大豆品种异黄酮主要组分含量。结果显示大豆籽粒中可检测出 6种主要的异黄酮组分 ,分别为大豆甙 (Daidzin)、甲氧基黄豆甙原 (Glycitin)、染料木甙 (Genistin)、丙二酰基大豆甙 (Malonyldaidzin)、丙二酰基黄豆甙原 (Malonylglycitin)和丙二酰基染料木甙 (Malonylgenistin)。各组分中以丙二酰基 (Mal onyl)异黄酮组分含量最高 (6 1 2 % ) ,且各组分间相关极显著。大豆品种间异黄酮含量变异较大 ,变异系数达 4 9 6 %。来自江苏省的品种海门红黄豆乙异黄酮含量最高 (4 932 3μg/ g) ,品种宝应等西风含量最低 (36 7 1μg/ g)。不同省份间异黄酮含量差异极显著 ,来自浙江省的大豆品种平均含量最高 (2 717 2 μg/ g) ,来自安徽省的平均含量最低 (1181 8μg/g)。异黄酮含量与生育期呈极显著正相关 (r=0 319 ) ,与百粒重呈显著正相关 (r=0 132 ) ,而与脂肪含量 (r =- 0 4 5 )和蛋白质含量 (r=-0 136 )呈负相关。 The contents of major isoflavone components in two hundred forty-nine soybean varieties from six southern provinces of China were analyzed by HPLC technology. Results showed that six major components of isoflavone could be determined by HPLC in soybean seeds, including daidzin, glycitin, genistin, malonyldaidzin, malonylglycitin and malonylgenistin. The component with the highest content was malonyl-isoflavone (61.2%), and correlations among components were statistically significant. Great variation among varieties was presented in 249 varieties, with the variation coefficient was 49.6%. Total isoflavone content of cultivar Haimenhonghuangdouyi was the highest (4932.3μg/g), that of cultivar Baoyingdengxifeng was the lowest (367.1μg/g). There was significant difference on isoflavone contents in different varieties among various provinces. The average content of isoflavone in Zhejiang province was the highest (2717.2μg/g), and the lowest content (1181.8μg/g) in Anhui province. There were a greatly significant positive correlation between isoflavone content and growth period (r=0.319 ***), a significant positive correlation between isoflavone content and weight of 100 seeds, and two negative correlations between isoflavone content and fat content (r= -0.45 ***) and protein content (r=-0.136) in soybean seeds.
