新型生物玻璃的细胞生物学评价

摘要 目的 对生物玻璃BG 2 0多孔支架进行细胞相容性研究。方法 采用L92 9小鼠成纤维细胞体外培养 ,按细胞增殖度法 ,取 7种浓度BG 2 0浸提液进行检测 ,分别于 2、4、6、8、10d用四唑盐 (MTT)比色法计数 ,计算细胞相对增殖度 ,用 6级毒性分类法评级 ;L92 9细胞接种于BG 2 0多孔支架体外培养 ,分别于 3d、7d取材 ,扫描电镜观察细胞在支架上的生长情况。结果 BG 2 0浸提液在高浓度时对细胞增殖有轻度抑制 ,低浓度对细胞增殖有一定的促进作用。BG 2 0毒性评级为 0~ 1级 ;扫描电镜示细胞在BG 2 0支架上粘附良好 ,形态正常。结论 BG 2 0作为一种新的多孔支架材料具有良好的细胞相容性。 Objective To evaluate the cytotoxicity of a novel porous bioglass BG-20 as scaffolds.Methods The proliferation of L929 mouse fibroblasts was investigated using a tetrazolium-based colorimetric assay(MTT assay).7 different concentrations of maceration extract of BG-20 were added into culture plates.The samples were tested at 2,4,6,8,10 days respectively after the addition of extract;L929 cells were seeded onto porous bioglass and the scaffolds in adhesion to fibroblasts were observed with scanning electron microscope(SEM)at 3 and 7 days.Results The maceration extract of BG-20 slightly inhibited L929 cell growth at the concentration enriched 8 times,however,lower concentrations promoted the multiplication of the cells.The toxicity gradation of BG-20 was 0~1.SEM observation revealed L929 cells seeded onto BG-20 scaffolds could adhere to the surface and maintain normal morphology.Conclusion As a novel scaffold material,BG-20 has satisfactory biocompatibility.
