摘要 目的 观察立体培养条件下牛晶状体前囊膜上皮细胞增殖情况。方法 将牛晶状体前囊膜用所设计的支架悬挂于无菌培养瓶中分别培养 1~ 6周 ,在不同培养时间用Giemsa染色后在倒置显微镜下观察细胞增殖情况。结果 所有前囊膜培养 3d后均有细胞增殖 ,并随时间推移增殖显著。 3周后增殖细胞开始脱落 ,6周后细胞全部脱落 ,囊膜上无细胞增殖。结论 在晶状体前囊膜立体培养时 ,晶状体上皮细胞的增殖和凋亡现象明显 ,向纤维细胞分化现象不明显。该培养模型可解释保留晶状体前囊膜白内障手术后的临床现象。 ObjectiveTo investigate the growth characteristics of bovine lens epithelial cells in vitro.MethodsThe bovine lens capsules were isolated and cultured in condition of5% CO 2 at 37 ℃by suspending with the special support for 6 weeks.In different time,the proliferation of the lens epithelial cells in the anterior capsule were observed by Giemsa staining and an inverted microscope.ResultsProliferation of lens epithelial cells were seen after 3 days. And the proliferating status of cells depended on the cultured time.After 3 weeks,the cells began to exfoliate and the lens capsule became transparent after 6 weeks.ConclusionThe proliferation and apoptosis of the lens epithelial cells are obvious in vitro three dimensional culture,but the differentiation toward fibroblasts is not considerable.This culture model can explain some clinical phenomenon caused by retaining the anterior capsule of lens during cataract surgery.
