摘要
建立了一种测定N 乙酰神经氨酸的在线富集毛细管区带电泳法 .该方法用 40mmol/L磷酸二氢钠作为缓冲溶液电解质 ,通过将样品溶解在乙腈∶水 ( 2∶1 )的样品基体中 ,进样时间增加到 1 0 0s来达到样品富集的效果 ,并考察了样品基体中乙腈浓度和进样时间对富集效果的影响 .研究了缓冲溶液pH值、三种不同电解质和有机修饰剂的加入对N 乙酰神经氨酸检测的影响 .最后考察了所建立方法的线性、检测限和重现性 ,并将该方法成功地应用于测定人体唾液中的N 乙酰神经氨酸 ,在 1 0min内完成分离 .
An on-line concentration capillary zone electrophoresis method was developed to determine N-acetyneuraminic acid in saliva.The influences of three different electrolytes,buffer pH and organic modifiers on the determination of N-acetyneuraminic acid were studied.The effects of the composition of sample matrix and injection time on the concentration efficiency of N-acetyneuraminic acid were also studied.Calibration line,detection limit and reproducibility of the developed method were investigated and the method was applied to determine N-acetyneuraminic acid in saliva.The separation was finished within 10*!min.
