摘要 建立检测人尿中兴奋剂 Carphedone分析方法。 5 0 m g尿样流经 XAD-2填装小柱淋洗 ,用甲醇洗脱后收集洗脱液 ,经过缓冲和酶解后振荡、离心 ,加入衍生化试剂 N-甲基 -N-三甲硅烷基三氟乙酰胺 ( MSTFA)和三甲硅烷基咪唑 ( TMSI)进行 TMS硅烷基化。气相色谱 -质谱 ( GC/MS)分析结果表明 :可以检测到 5 0 h内的代谢产物 ,同时可以检测 Carphedone衍生化后的脱水反应产物。 Carphedone(2-Oxo-4-phenylpyrrolidine acetamide) as a new stimulant has been banned by International Olympic Committee(IOC) since 1997. After administration of single dose of the drug,the 50mg of urine samples were extracted by use of XAD packed minicolumn and eluted with methanol. The concentrate solution was collected, buffered, enzymed, centrifuged and derivated with N-methyl-N-trimethylsilyl trifluoroacetamide(MSTFA) under the condition of procedure for anabolic agents as the routine method in the lab. Carphedone itself was detected by gas chromatography-mass spectrometry(GC/MS). The results showed the metabolization products within 50 hours and dehydrated products of the molecule are found and recognized to be formed during derivatization.
