摘要 背景:注意缺陷多动障碍(attentiondeficithyperactivitydisorder,ADHD)存在着强化机制的异常,而多动性是强化机制异常的一种表现。目的:观察ADHD动物模型自发高血压大鼠(SHR)的多动性变化,以探究其强化机制。设计:非随机对照的实验研究。地点、材料和干预:实验在挪威奥斯陆大学医学系生理实验室进行。实验选用12只雄性SHR大鼠和12只雄性WistarKyoto(WKY)大鼠。实验在标准Campden连续反应测试箱中进行,大鼠禁水22h,采用长变异间期方案(变异指数180s),分析最后10d稳定曲线的均值。主要观察指标:大鼠按杠次数、推门次数及在不同阶段的变化。结果:①SHR组大鼠按杠次数犤(642.9±173.1)次犦显著多于WKY组犤(219.9±79.5)次〗(t=7.52,P<0.01)。②实验过程的3个相同阶段SHR大鼠的按杠次数比较,差异无显著性意义(F=0.10,P>0.05)。③SHR组的推门次数明显多于WKY组(t=4.43,P<0.01),而获水次数与WKY组比较,差异无显著性意义(t=1.86,P>0.05)。结论:SHR大鼠的多动性较WKY大鼠强,强化机制异常,有助于了解ADHD的发病机制、临床治疗及预防。 BACKGROUND:Attention deficit hyperactivity disorder(ADHD) has abnormal strengthening mechanism and hyperactivity is one of the expressions of this abnormal strengthening mechanism. OBJECTIVE:To investigate the behavioral variations of spontaneous hypertensive rat(SHR),an ADHD animal model,for the exploration of its strengthening mechanism. DESIGN:A non randomized controlled trial. SETTING,PARTICIPANTS and INTERVENTIONS:The study was conducted in the Laboratory of Physiology of the Faculty of Medicine of the University of Oslo,Norway.12 male SHR rats and 12 male Wistar Kyoto(WKY) rats were selected.The experiment was carried on in the standard Campden instrument operant chambers.The animals were water deprived for 22 hours.A long Variable Interval Schedule(variable index:180 s) was used to analyze the mean value of the stable curve of the last ten days. MAIN OUTCOME MEASURES:The times of lever pressing and door opening and the variation at different stages. RESULTS:①The lever pressing time of SHR group(642.9±173.1) was significantly more than that of WKY group[(219.9±79.5),t=7.52,P< 0.01].②There were no significant differences of bar pressing among three different stages(F=0.10,P >0.05). ③The door opening times of SHR group was significantly more than that of WKY group(t=4.43,P< 0.01);however,the times of water obtaining was not significantly different from that of WKY group(t=1.86,P >0.05). CONCLUSION:The hyperactivity of SHR is much more obvious than that of the WKY and it has abnormal strengthening mechanism,which is helpful for the investigation of the pathogenesis and clinical therapy and prevention of ADHD.
