皖籍汉族人寻常型银屑病患者TNF-α基因多态性分析 被引量:5

Association of tumor necrosis factor-alpha gene promoter polymorphism with psoriasis vulgaris in the Han population

摘要 目的 探讨TNF α基因多态性与皖籍汉族人寻常型银屑病的相关性。方法 采用PCR RFLP方法 ,检测皖籍汉族人 6 5例早发型 (Ⅰ型 )银屑病、2 2例迟发型 (Ⅱ型 )银屑病及 1 2 8例健康人TNFα- 2 38多态性。结果 早发型银屑病患者携带TNFα - 2 38.2 (TNF G/A基因型 )频率较对照组明显增高 (2 6 .2 %vs 1 0 .9% ) ,二者差异有显著性 (OR =2 .88,Pc<0 .0 5 ) ;而迟发型银屑病患者携带该基因型频率则与对照组差异无显著性 (9.1 %vs 1 0 .9% ,OR =0 .81 ,Pc>0 .0 5 )。但早发型银屑病男女患者携带TNFα- 2 38.2频率与相应对照组差异均无显著性 (Pc>0 .0 5 )。结论 TNFα- 2 38.2频率与皖籍汉族人寻常型银屑病发病类型明显关联 ,提示TNF α基因多态性可能为汉族人银屑病易感因子之一。 Objective To investigate the association of tumor necrosis factor alpha (TNFα) gene promoter polymorphism with psoriasis vulgaris (PV) in the Han population of Anhui province. Methods Using PCR RFLP (restriction fragment length polymorphism), we detected the frequency of TNFα gene promoter polymorphism in locus -238 among 65 patients with early onset psoriasis (type Ⅰ), 22 patients with late onset psoriasis (type Ⅱ), and 128 unrelated healthy controls in the Hans, respectively. Results The distributing frequency of TNF G/A heterozygous genotype (TNFα-238.2) in locus-238 increased significantly in early onset patients with PV (26.2% vs 10.9% , OR=2.88 , P c <0.05 ), not in late onset patients, comparing with the controls. No difference was found in the frequency of TNFα-238.2 in male and female patients with early onset psoriasis. Conclusions TNFα-238 polymorphism was highly associated with the type Ⅰ psoriasis, which indicated that TNFα gene polymorphism may be one of genetic susceptibility factors of psoriasis in Han population.
