摘要
①目的 探讨基质金属蛋白酶 9(MMP 9)和转化生长因子 β1 (TGF β1 )在原发性高血压 (HP)、高血压肾小动脉硬化 (HN)发生发展中的作用。②方法 采用逆转录聚合酶链反应 (RT PCR)技术 ,分别检测HP、HN和肾小动脉硬化肾衰竭 (HNF)病人外周血单个核细胞中MMP 9、TGF β1 mRNA表达。③结果 HN及HNF组MMP 9mRNA表达水平较正常组 (NC)显著增高 (F =1 37.70 ,q =1 4 .5 7、2 3.37,P <0 .0 0 1 ) ;HP、HN、HNF 3组TGF β1 mRNA表达水平较NC组均显著增高 (F =1 0 3.6 8,q =4 .4 7~ 2 2 .97,P <0 .0 1 )。 ④结论 MMP 9、TGF β1可能介导了高血压动脉硬化、小动脉玻璃样变及血管重塑过程。在高血压进展过程中TGF β1 早期表达 ,并上调MMP 9的表达 。
Objective To investigate the roles of matrix metalloproteinase-9(MMP-9) and transform growth factor-β 1(TGF-β 1) in the development and progress of primary hypertension (HP) and hypertensive renal arteriolar sclerosis(HRAS). Methods MMP-9 and TGF-β 1 expressions were detected by reverse transcription-PCR(RT-PCR) in peripheral blood monocytes of patients with PH, HRAS and renal arteriolar sclerosis renal failure(RASRF). Results A significantly higher level of MMP-9 mRNA was detected in the HRAS and RASRF groups than in the healthy controls ( F=137.70; q=14.57,23.37;P <0.001). The level of TGF-β 1 mRNA was also higher in the PH, HRAS and RASRF groups than that of the controls ( F=103.68,q=4.47- 22.97 , P <0.01). Conclusion MMP-9 and TGF-β 1 may mediate hypertensive arteriosclerosis, hyaline change of arterioles, and vascular remodeling. The early expression of TGF-β 1 and the up-regulated expression of MMP-9 accelerate the renal injuries.
