Anti-HBs Ag单克隆抗体免疫吸附剂的制备及评价

摘要  以环氧氯丙烷、脱乙酰几丁质和NaOH为原料,在活化温度43℃下制得含有活化基团的中间体,再固定Anti HBsAg单克隆抗体,得到偶联率为18.7%的Anti HBsAg单克隆抗体免疫吸附剂.经患者血清体外吸附评价实验可知:该吸附剂可吸附除去30%~50%致病抗体,血清由阳性转为阴性. In order to remove the HBsAg(Hepatitis B surface antigen) from the blood of patients with hepatitis B, the immunoadsorbent was prepared by coupling Anti-HBsAg monoclonal antibody to epichlorohydrin-activated chitosan resins. The methods and conditions for activation of carrier, Anti-HBsAg monoclonal antibody coupling and adsorption with patient's plasma were investigated. The adsorption tests showed that the immunoadsorbent could remove 30 %~50 % of HBsAg from the plasma when coupling rate of epichlorohydrin-activated chitosan resins to Anti-HbsAg monoclonal antibody was 18.7 %.
