摘要
用电化学方法和UV光谱法对Pd(Ⅱ) 5 Br PADAP络合物与鱼精子DNA的相互作用进行了研究,发现在pH6.5的六次甲基四胺缓冲溶液中,Pd(Ⅱ) 5 Br PADAP络合物与鱼精子DNA以嵌入方式结合生成了非电活性的超分子化合物,导致络合物的峰电流降低,峰电流值在一定范围内与DNA的质量浓度成线性关系,线性范围为0 3~3μg/mL,检出限为0 02μg/mL,相关系数为0 998。方法可以用于DNA的测定。
The interaction of Pd(Ⅱ)-5-Br-PADAP (2-(5-bromo-2-pyridylazo)-5-diethylaminopheno) with herring sperm DNA was studied by electrochemical and UV-spectrophotometric methods. Pd(Ⅱ)-5-Br-PADAP reacted with herring sperm DNA in a mode of intercalation to form an electrochemically non-active super molecular complex in the buffer solution of (CH_2)_6N_4 (pH 6.5), resulting in the decrease of the peak current of Pd(Ⅱ)-5-Br-PADAP. The peak current decrease was proportional to the concentration of DNA from 0.3~3 μg/mL with the correlation coefficient of 0.998 and the detection limit of 0.02 μg/mL.
